Abstract

Translationally controlled tumor protein, also known as fortilin, is a 172 amino acid cytosolic shuttle protein of 20 kDa that prevents apoptosis through various mechanisms and accelerates atherosclerosis by allowing macrophage propagation. The goal of this research is to express 15N/13C labeled fortilin in E. coli cells and use novel small molecular weight (SMW) inhibitors that are derived from previously determined fortilin inhibiting small molecular weight compounds (FISCs) and evaluate their potency by determining their ability to bind and inhibit fortilin using a combination of computational methods, nuclear magnetic resonance (NMR) and surface plasmon resonance (SPR).
