Abstract

Sphingolipids constitute major building blocks of membranous organelles, lipid rafts and external environment barriers. This poster focuses on how sphingolipids are an important part of the physiological process and a basic building block of a lipid membrane. Jaspine B, anhydrophytosphingosine – sphingolipid mimic, has exhibited mitochondrial membrane depolarization as verified by cytochrome c release and apoptosis induction. Spisulosine, a sphingoid mimic was synthesized utilizing chiral pool strategy using L-amino acid. A stereoselective synthesis was developed utilizing L-serine. The total synthesis involved development of a second generation chiral intermediate “Aminobutenolide” – Dutta Lactone followed by FGI, intramolecular cyclization, and organo-metallic reactions. The structural similarity of Jaspine B with sphingolipid, jaspine B has evolved as probe to study the membrane organelle function, for example, mitochondrial function. The invitro studies, involving the NCI-60 panel screening of sphingolipid mimics will be discussed.
